Check your windows firewall.

Start > Control Panel > Network connections

Right click your network connection you use, goto properties on the popup

New window, goto Advanced Tab at the top, settings for the firewall.

Turn it off and check to see if it connects.

Failing that, check with the publisher or developers website for the game and check the FAQ's they have for online play.
